      Description
      • Sorting-related receptor with A-type repeats (SORLA) is an intracellular sorting receptor that directs cargo proteins, such as kinases, phosphatases, and signaling receptors, to their correct location within the cell. SORL1 functions as a suppressor of Aβ secretion through its dual binding to both APP and Aβ.
      • The amino acid at position 528 is mutated from alanine (Ala) to threonine (Thr) in B-Sorl1*A528T mice.
      • Mouse Sorl1 mRNA was both detectable in wild-type and heterozygous B-Sorl1*A528T mice, and the point mutation was confirmed via Sanger sequencing.
      • This model is useful for studying the mechanisms of SORL1 point mutations in Alzheimer’s disease (AD).

      mRNA Expression Analysis

      Strain specific analysis of Sorl1 mRNA expression in wild-type C57BL/6JNifdc mice and B-Sorl1*A528T mice by RT-PCR. Cortex RNA were isolated from wild-type C57BL/6JNifdc mice (+/+) and heterozygous B-Sorl1*A528T mice (Mut/+), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse Sorl1 primers. Mouse Sorl1 mRNA was both detectable in wild-type and heterozygous B-Sorl1*A528T mice, and the point mutation was confirmed via Sanger sequencing.
