      Description
      • Tumor Necrosis Factor (TNF), a pleiotropic pro-inflammatory cytokine belonging to the TNF superfamily, is involved in the regulation of diverse biological processes. The AU-rich element (ARE) located within the 3' UTR of Tnfα destabilizes mRNA under steady-state conditions.
      • In B-Tnf△ARE mice, the deletion of this ARE sequence enhances the stability of Tnfα mRNA, leading to chronically elevated levels of TNF protein.
      • The ARE sequence in the 3'UTR of the mouse Tnf gene was deleted via gene editing, while all other sequences remained unchanged.
      • Under SPF housing conditions, the serum TNF protein level in B-Tnf△ARE mice is relatively low, at approximately 3 pg/mL, whereas no expression is detectable in wild‑type mice.
      • Under LPS-induced conditions, serum TNF protein levels were elevated in both B-Tnf△ARE mice and wild-type (WT) mice; however, the protein levels in B-Tnf△ARE mice were significantly higher than those in the WT group. Over time, the TNF expression in WT mice rapidly returned to basal levels, whereas B-Tnf△ARE mice maintained sustained high levels for a prolonged period.
      • This product model is expected to serve as a valuable tool for research into ileitis and arthritis, functioning as a robust disease model.
      • Homozygous mice are expected to exhibit severe phenotypes, resulting in embryonic lethality or poor postnatal survival; heterozygotes are suitable for research applications

      Protein expression analysis in serum

      Mouse TNF expression analysis in C57BL/6JNifdc mice and heterozygous  B-Tnf△ARE mice by ELISA. Serum was collected from C57BL/6JNifdc mice and heterozygous  B-Tnf△ARE mice (Female, n=3). Expression level of mouse TNF were analyzed by ELISA (mouse TNF, Biolegend, 430904). Mouse TNF was detectable in heterozygous  B-Tnf△ARE mice. Values are expressed as mean ± SEM.

      Mouse TNF expression analysis in C57BL/6JNifdc mice and heterozygous  B-Tnf△ARE mice by ELISA. Serum was collected from C57BL/6JNifdc mice and heterozygous  B-Tnf△ARE mice (Female, n=3), 1h, 3h, and 6h after LPS stimulation (20 ug/200 ul. i.p.). Expression level of mouse TNF were analyzed by ELISA (mouse TNF, Biolegend, 430907). Mouse TNF was detectable in heterozygous  B-Tnf△ARE mice.
