Targeting strategy
Gene targeting strategy for B-hIL19 mice. The exons 2-6 of mouse Il19 gene that encode the full coding sequences were replaced by human IL19 exons 2-6 in B-hIL19 mice.
mRNA expression analysis
Strain specific analysis of IL19 gene expression in wild-type C57BL/6 mice and B-hIL19 mice by RT-PCR. Mouse Il19 mRNA was detectable in spleen of wild-type C57BL/6 mice (+/+) but not in homozygous B-hIL19 mice (H/H). Human IL19 mRNA was detectable only in homozygous B-hIL19 mice but not in wild-type mice.
Protein expression analysis
Strain specific IL19 expression analysis in homozygous B-hIL19 mice by ELISA. Plasma were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hIL19 mice (H/H) stimulated with or without LPS in vivo and analyzed by ELISA with strain-specific anti-IL19 ELISA kit. Mouse IL19 was only detectable in wild-type mice. Human IL19 was only detectable in homozygous B-hIL19 mice.
