- ITGA10 is a 160 kDa member of the integrin alpha chain family of molecules. Mature human integrin alpha 10 is a 1145 amino acid (aa) type I transmembrane glycoprotein that contains a 1100 aa extracellular domain (ECD) (aa 23-1122) plus a 22 aa cytoplasmic tail. The ECD contains two FG-GAP regions (aa 24-95), followed by one vWFA domain (aa167-350) and five consecutive FG-GAP regions (aa 361-657). The FG-GAP repeats are suggested to participate in the formation of a beta -propeller. ITGA10 is expressed at high levels in chondrocytes, where it is transcriptionally regulated by AP-2epsilon and Ets-1. Diseases associated with ITGA10 include Myxofibrosarcoma and Spinal Muscular Atrophy, Type Iv. Among its related pathways are Apoptotic Pathways in Synovial Fibroblasts and Integrin Pathway. Gene Ontology (GO) annotations related to this gene include collagen binding and collagen binding involved in cell-matrix adhesion. ITGA10, along with integrin beta 1, is thought to be an essential part of a novel collagen type II receptor expressed mainly in chondrocytes and cartilage tissue.

Targeting strategy
Gene targeting strategy for B-hITGA10 mice.
The exons 2~29 of mouse Itga10 gene that encode the extracellular domain were replaced by human ITGA10 exons 2~29 in B-hITGA10 mice.
The genomic region of mouse Itga10 gene that encodes signal peptide and cytoplasmic portion is retained. The promoter, 5’UTR and 3’UTR region of the mouse gene are also retained.
The chimeric Itga10 expression is driven by endogenous mouse Itga10 promoter, while mouse Itga10 gene transcription and translation will be disrupted.
mRNA expression analysis
Strain specific analysis of ITGA10 mRNA expression in wild-type C57BL/6JNifdc mice and B-hITGA10 mice by RT-PCR. Lung mRNA were isolated from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hITGA10 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human ITGA10 primers. Mouse Itga10 was detectable only in wild-type mice. Human ITGA10 mRNA was detectable only in homozygous B-hITGA10 mice but not in wild-type mice.
Protein expression analysis
Western blot analysis of ITGA10 protein expression in homozygous B-hITGA10 mice. Various tissue lysates were collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hITGA10 mice (H/H), and then analyzed by western blot with anti-ITGA10 antibody antibody (Thermo Fisher, PA5-67829 , cross-reactive between human and mouse).
