      • FZD10 is a member of the frizzled gene family, encodes 7-transmembrane domain proteins that are receptors for the Wingless type MMTV integration site family of signaling proteins. Acts upstream of or within Wnt signaling pathway. Is expressed in several structures, including alimentary system; central nervous system; genitourinary system; integumental system; and sensory organ.
      • Mouse Fzd10 gene only one exon, part of this exon that encode the whole molecule (ATG to STOP codon) were replaced by human counterparts in B-hFZD10 mice.
      • Mouse Fzd10 mRNA was only detectable in wild-type mice. Human FZD10 mRNA was exclusively detectable in homozygous B-hFZD10 mice but not in wild-type mice.
      Targeting strategy

      Gene targeting strategy for B-hFZD10 mice.

      Mouse Fzd10 gene only one exon, part of this exon that encode the whole molecule (ATG to STOP codon) were replaced by human counterparts in B-hFZD10 mice.

      The promoter and 5’UTR region of the mouse gene are retained, and the mouse 3’UTR is replaced by the human 3’UTR.

      The human FZD10 expression was driven by endogenous mouse Fzd10 promoter, while mouse Fzd10 gene transcription and translation will be disrupted.

      mRNA expression analysis

      Strain specific analysis of FZD10 mRNA expression in wild-type C57BL/6 mice and B-hFZD10 mice by RT-PCR. Cerebellum RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hFZD10 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human FZD10 primers. Mouse Fzd10 mRNA was detectable only in wild-type C57BL/6 mice. Human FZD10 mRNA was detectable only in homozygous B-hFZD10 mice but not in wild-type mice.
