      Description
      • The major inhibitory neurotransmitter in the mammalian brain, γ-aminobutyric acid (GABA), is synthesized by two glutamic acid decarboxylase (GAD) isoforms, GAD65 and GAD57. GAD65 is localized to the nerve terminal and is reversibly bound to the membrane of synaptic vesicles.
      • The P2A-iCre-T2A-Antares expression cassette was in-frame inserted immediately after Leu585 codon in mouse Gad2 gene in B-Gad2-Cre-Antares mice.
      • To validate the Cre recombinase activity, B-Gad2-Cre-Antares mice were crossed with CAG-tdTomato reporter mice, utilizing tdTomato expression as an indicator of Cre recombinase activity. Detectable tdTomato expression was observed, confirming the Cre recombinase was functional.
      • This product is used for labeling and lineage tracing of GAD2-positive cells.
      Targeting Strategy

      Gene targeting strategy for B-Gad2-Cre-Antares mice. The P2A-iCre-T2A-Antares expression cassette was in-frame inserted immediately after Leu585 codon in mouse Gad2 gene in B-Gad2-Cre-Antares mice.

      tdTomato Expression Analysis

      Representative histological images of brain from B-Gad2-Cre-Antares mice and B-Gad2-Cre-Antares, CAG-tdTomato cKI mice. The whole brain sections from 9-week-old heterozygous B-Gad2-Cre-tdTomato mice (Mut/+) and double heterozygous B-Gad2-Cre-Antares, CAG-tdTomato cKI mice (Mut/+, Mut/+) were presented to display the expression of tdTomato. tdTomato was assay by IHC using tdTomato antibody (HUABIO, HA500049). tdTomato were only detectable in hippocampus, cortex and cerebellum in B-Gad2-Cre-Antares, CAG-tdTomato cKI mice, but not in B-Gad2-Cre-Antares mice. Scare bar is 50μm.
