      Description
      • CD157, also known as bone marrow stromal cell antigen 1 (BST-1), is a glycosyl phosphatidylinositol anchored membrane protein that belongs to the CD38 family, which may play a role in rheumatoid arthritis (RA) due to its enhanced expression in RA‑derived bone marrow stromal cell lines.
      • The exons 1-9 of mouse Bst1 gene that encode the whole molecule (ATG to STOP codon) were replaced by human counterparts in B-hBST1 mice. The promoter, 5’UTR and 3’UTR region of the mouse gene are retained. The human BST1 expression is driven by endogenous mouse Bst1 promoter, while mouse Bst1 gene transcription and translation will be disrupted.
      • Mouse Bst1 mRNA was detectable in wild-type C57BL/6 mice. Human BST1 mRNA was detectable in homozygous B-hBST1 mice.
      • Human BST1 was detectable in monocytes、macrophages、DCs、neutrophils from spleen and blood of B-hBST1 mice.
      • Application: This product is used for pharmacodynamics and safety evaluation of drugs.

      mRNA expression analysis in humanized B-hBST1 mice

      Strain specific analysis of BST1 mRNA expression in wild-type C57BL/6JNifdc and B-hBST1 mice by RT-PCR. Spleen RNA was isolated from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hBST1 mice (H/H). Mouse Bst1 mRNA was detectable only in wild-type C57BL/6JNifdc mice. Human BST1 mRNA was detectable only in homozygous B-hBST1 mice, but not in wild-type C57BL/6JNifdc mice.

      Protein expression analysis-Spleen

      Strain specific BST1 expression analysis in homozygous B-hBST1 mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hBST1 mice (H/H) and analyzed by flow cytometry with species-specific anti-BST1 antibody(eBioscience™, 12-1579-42). hBST1 was detectable in T cells (weak expression), B cells(weak expression), monocytes and macrophages of homozygous B-hBST1 mice, but not in wild-type mice.
