      Targeting strategy

      Gene targeting strategy for B-hCCR2 mice. The exons 3 of mouse Ccr2 gene that encode the full-length protein domain was replaced by human CCR2 exons 2-3 in B-hCCR2 mice.

      Protein expression analysis

      Strain specific CCR2 expression analysis in homozygous B-hCCR2 mice by flow cytometry. Bone marrow was collected from WT and homozygous B-hCCR2 mice, and analyzed by flow cytometry with species-specific anti-CCR2 antibody. Mouse CCR2 was only detectable in WT mice. Human CCR2 was exclusively detectable in homozygous B-hCCR2 but not WT mice.

      Evaluation of the CCR2+ Monocyte Depleting Agent in B-hCCR2 mice

      Evaluation of CCR2+ Monocyte Depleting Agent in B-hCCR2 mice. B-hCCR2 mice (8-week-old, female) were randomly enrolled into 2 study groups (n=4 per group) based on body weight. The test article was administered on the day of grouping (Day 0). 24 hours after dosing, anticoagulated blood was collected and analyzed by flow cytometry. The test article reduced the monocyte percentage and the human CCR2-positive monocyte percentage in B-hCCR2 mice (CD192 is the alias for CCR2). Data were shown as Mean±SEM, and analyzed by a t-test.
