      Description

CD40 is a member of the tumor necrosis factor receptor (TNFR) superfamily and is primarily expressed on antigen-presenting cells, including dendritic cells, B cells, macrophages, and monocytes. Interaction between CD40 and its ligand CD40L (CD154) plays a central role in adaptive immunity by promoting antigen presentation, cytokine production, and T-cell priming. Due to its immunostimulatory function, CD40 is a key target for cancer immunotherapy, vaccine adjuvant strategies, and immune modulation research.

In CD40 humanized mice (B-hCD40), the endogenous murine Cd40 gene is replaced with the human CD40 coding sequence to enable species-specific expression of human CD40 under native regulatory control. This humanization design supports translationally relevant evaluation of anti-human CD40 biologics by allowing therapeutic engagement with human CD40 in an in vivo immunocompetent setting while eliminating interference from murine CD40.

Validation data confirm the successful establishment of CD40 humanized mice (B-hCD40), supporting their use as a preclinical model for pharmacodynamic, efficacy, and safety assessment of CD40-targeting therapeutics.

      Key Advantages

      • Species-specific human CD40 expression
Human CD40 is specifically expressed in CD40 humanized mice (B-hCD40), enabling accurate in vivo evaluation of human CD40-targeting antibodies without confounding murine CD40 activity.
      • Translational platform for anti-human CD40 antibody development
CD40 humanized mice (B-hCD40) provide a practical preclinical model to study CD40 pathway modulation and support pharmacodynamic assessment of agonistic or antagonistic anti-human CD40 biologics.
      • Supports integrated efficacy and safety assessment
By enabling human CD40 engagement in an immunocompetent background, CD40 humanized mice (B-hCD40) support combined readouts including immune activation, tumor immune profiling, and safety-relevant endpoints for candidate selection and dose optimization.

      Validation

      • Human CD40 expression:
Flow cytometry and molecular analyses confirmed that human CD40 is specifically and robustly expressed in homozygous CD40 humanized mice (B-hCD40), while endogenous mouse Cd40 expression is absent. In contrast, wild-type C57BL/6 mice express only murine CD40, demonstrating successful and species-specific human CD40 replacement.
      • Functional CD40 signaling and immune activation:
CD40 humanized mice (B-hCD40) exhibited intact and functional human CD40–dependent immune responses, including efficient activation of antigen-presenting cells following stimulation with anti-human CD40 agonistic antibodies, as evidenced by upregulation of costimulatory molecules and downstream immune activation.
      • Therapeutic relevance for CD40-targeted immunotherapy:
Treatment with anti-human CD40 antibodies in CD40 humanized mice (B-hCD40) resulted in robust in vivo antitumor immune modulation, supporting the model’s suitability for preclinical evaluation of CD40-targeting antibodies, immune agonists, and combination immunotherapy strategies.

      Application

CD40 humanized mice (B-hCD40) are used for in vivo evaluation of CD40-targeted antibodies, immune agonists, and combination immunotherapies by enabling human CD40-specific immune activation, antitumor efficacy assessment, and translational safety profiling.

Targeting Strategy for CD40 humanized mice

      CD40

      • The exons 2-7 of mouse Cd40 gene that  encode the extracellular domain were replaced by human CD40 exons 2-7 in CD40 humanized mice (B-hCD40).
      • The endogenous mouse promoter, 5′ UTR, and 3′ UTR regions are retained, allowing human CD40 expression to be driven by the native mouse Cd40 promoter, while endogenous mouse Cd40 transcription and translation are abolished.
