      Description
      • CLEC6A (C-type lectin domain containing 6A), also known as dectin-2, belongs to the C-type lectin family of transmembrane immune regulatory glycoproteins. The encoded protein is a type II membrane receptor with an extracellular C-type lectin-like domain fold. When stimulated, the encoded protein initiates signaling through the CARD9-Bcl10-Malt1 pathway, leading to the induction of cytokines.
      • The exons 3-6 of mouse Clec6a gene that encode extracellular domain is replaced by human counterparts in B-hCLEC6A mice. The genomic region of mouse Clec6a gene that encodes cytoplasmic portion and transmembrane domain is retained. The promoter, 5’UTR and 3’UTR region of the mouse gene are also retained. The CLEC6A expression is driven by endogenous mouse Clec6a promoter, while mouse Clec6a gene transcription and translation will be disrupted.
      • Mouse Clec6a mRNA was detectable in wild-type C57BL/6N mice. Human CLEC6A mRNA was detectable in homozygous B-hCLEC6A mice.
      • Human CLEC6A was detectable in DCs, neutrophils, and macrophages of homozygous humanized B-hCLEC6A mice but not in wild-type C57BL/6N mice.
      • Application: This product is used for preclinical pharmacological evaluation of antibody drugs.
      Targeting strategy

      Gene targeting strategy for B-hCLEC6A mice. The exons 3-6 of mouse Clec6a gene that encode extracellular domain is replaced by human counterparts in B-hCLEC6A mice. The genomic region of mouse Clec6a gene that encodes cytoplasmic portion and transmembrane domain is retained. The promoter, 5’UTR and 3’UTR region of the mouse gene are also retained.       Protein expression analysis-Spleen

      Strain specific CLEC6A expression analysis in homozygous B-hCLEC6A mice by flow cytometry. Spleen cells were collected from wild-type C57BL/6N (+/+) and homozygous B-hCLEC6A mice (H/H), and analyzed by flow cytometry with species-specific anti-hCLEC6A antibody (Human Dectin-2/CLEC6A Antibody, RD, MAB31141). Human CLEC6A was detectable in  DCs and macrophages of B-hCLEC6A mice.

      mRNA expression analysis

      Strain specific analysis of CLEC6A mRNA expression in wild-type C57BL/6N mice and B-hCLEC6A mice by RT-PCR. Lung RNA was isolated from wild-type C57BL/6N mice (+/+) and homozygous B-hCLEC6A mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human CLEC6A primers. Mouse Clec6a mRNA was only detectable in wild-type mice. Human CLEC6A mRNA was exclusively detectable in homozygous B-hCLEC6A mice but not in wild-type mice.
