Gene targeting strategy for B-hCLEC6A mice. The exons 3-6 of mouse Clec6a gene that encode extracellular domain is replaced by human counterparts in B-hCLEC6A mice. The genomic region of mouse Clec6a gene that encodes cytoplasmic portion and transmembrane domain is retained. The promoter, 5’UTR and 3’UTR region of the mouse gene are also retained. The CLEC6A expression is driven by endogenous mouse Clec6a promoter, while mouse Clec6a gene transcription and translation will be disrupted.
Strain specific CLEC6A expression analysis in homozygous B-hCLEC6A mice by flow cytometry. Spleen cells were collected from wild-type C57BL/6N (+/+) and homozygous B-hCLEC6A mice (H/H), and analyzed by flow cytometry with species-specific anti-hCLEC6A antibody (Human Dectin-2/CLEC6A Antibody, RD, MAB31141). Human CLEC6A was detectable in DCs and macrophages of B-hCLEC6A mice.
Strain specific analysis of CLEC6A mRNA expression in wild-type C57BL/6N mice and B-hCLEC6A mice by RT-PCR. Lung RNA was isolated from wild-type C57BL/6N mice (+/+) and homozygous B-hCLEC6A mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human CLEC6A primers. Mouse Clec6a mRNA was only detectable in wild-type mice. Human CLEC6A mRNA was exclusively detectable in homozygous B-hCLEC6A mice but not in wild-type mice.
