      Description

GLP1R (Glucagon-like peptide-1 receptor) is a G protein–coupled receptor (GPCR) highly expressed in pancreatic β-cells and critical for glucose-dependent insulin secretion, appetite regulation, and energy balance. GLP1R is the therapeutic target of GLP-1 receptor agonists (GLP-1RAs), such as semaglutide and liraglutide, which are widely used for the treatment of type 2 diabetes and obesity.

In GLP1R humanized mice, the murine Glp1r gene is replaced with the human GLP1R gene. Homozygous mice express human GLP1R in physiologically relevant tissues without altering immune or metabolic homeostasis. This model provides a translationally relevant platform for preclinical efficacy testing, antibody validation, and drug development targeting GLP1R.

GLP1R humanized mice are an ideal preclinical model for evaluating GLP1R-targeted therapeutics in metabolic disease research, including type 2 diabetes and obesity.

      Key Advantages

      • Complete human gene replacement:
Murine Glp1r replaced by human GLP1R.
      • Physiological expression:
Human GLP1R detected in pancreas and gut, mimicking human metabolic regulation.
      • Drug response fidelity:
Accurately models pharmacological response to GLP-1 receptor agonists.
      • Stable baseline phenotype:
No disruption of immune or metabolic balance.
      • Preclinical translational relevance:
Directly applicable to diabetes and obesity drug development.
      • Unique Biocytogen advantage:
Full humanization strategy ensures superior predictive power compared with transgenic or partial replacement models.

      Validation

      • Human GLP1R expression confirmed in homozygous GLP1R humanized mice by RT-PCR and Western blot.
      • Protein localization: Human GLP1R detected in pancreatic islets and intestinal L-cells, consistent with physiological expression patterns.
      • Functional assays: GLP1R humanized mice respond to GLP-1 analogs (exendin-4, liraglutide) with glucose-lowering effects comparable to human physiology.
      • Metabolic phenotype: Homozygous GLP1R humanized mice exhibit normal body weight, glucose tolerance, and insulin sensitivity compared to wild-type controls.

      Applications

      • Diabetes Research:
Test efficacy of GLP-1 receptor agonists and novel incretin-based therapies.
      • Obesity & Metabolic Syndrome Studies:
Evaluate effects of GLP1R-targeted drugs on appetite, energy expenditure, and weight regulation.
      • Antibody Validation:
Assess binding, efficacy, and safety of anti-human GLP1R antibodies in vivo.
      • Drug Discovery & Development:
Screen small molecules, peptides, and biologics targeting GLP1R.
      • Mechanistic Studies:
Explore GLP1R-mediated pathways in glucose metabolism, cardiovascular protection, and gastrointestinal regulation.
      Targeting strategy

      GLP1R

      • The full coding sequence of the human GLP1R gene was inserted into the mouse exon 1 of the Glp1r gene in GLP1R humanized mice (B-hGLP1R). The human GLP1R protein expression will be driven by the endogenous mouse Glp1r promoter, while mouse Glp1r gene transcription and translation will be disrupted.
