      Description
      • Galectin-10 (also eosinophil lysophospholipase and Charcot-Leyden Crystal protein) is a 16 kDa member of the lectin family of proteins. It is expressed intracellularly by eosinophils, basophils and CD25+ Treg cells. It is known to bind lysophospholipase and its inhibitors and to bind mannose in a very unusual manner. There is no known structural rodent counterpart to human Galectin-10.
      • The full coding sequences of human LGALS10 gene, including the promoter, 5’UTR and 3’UTR are inserted into mouse Hipp11 (H11) locus in B-hLGALS10 mice.
      • Human LGALS10 mRNA was detectable only in homozygous B-hLGALS10 mice but not in wild-type mice.
      • LGALS10 were detectable in bone marrow, gall bladder, colon, stomach and spleen in homozygous B-H11-hLGALS10 mice but not in wild-type mice.
      • B-hLGALS10 mice can be used to study the in vivo efficacy evaluation of LGALS10 antibody drugs.
      Targeting strategy

      Gene targeting strategy for B-hLGALS10 mice. The full coding sequences of human LGALS10 gene, including the promoter, 5’UTR and 3’UTR are inserted into mouse Hipp11 (H11) locus in B-hLGALS10 mice.

      mRNA expression analysis

      Strain specific analysis of LGALS10 mRNA expression in wild-type C57BL/6 mice and B-hLGALS10 mice by RT-PCR. Spleen RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hLGALS10 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with human LGALS10 primers. Human LGALS10 mRNA was detectable only in homozygous B-hLGALS10 mice but not in wild-type mice.

      Protein expression analysis

      Western blot analysis of LGALS10 protein expression in homozygous B-hLGALS10 mice. Various tissue lysates were collected from wide type mice and homozygous B-hLGALS10 mice (H/H), and then analyzed by western blot with anti-LGALS10 antibody (R&D, AF5447). 40 μg total proteins were loaded for western blotting analysis. LGALS10 were detectable in bone marrow, gall bladder, colon, stomach and spleen in homozygous B-hLGALS10 mice but not in wild-type mice.

      (Note: The beta-actin of gall bladder in wild-type mice was presumed to have degraded.)
