      CD8 and CD20-Based Therapy 

      • Gene Information: CD8: Encoded by CD8A/CD8B on chromosome 2p11.2, functioning as a vital cytotoxic T-cell co-receptor. CD20: Encoded by MS4A1 on chromosome 11q12.2, a four-transmembrane phosphoprotein regulating B-cell activation and calcium influx. 
      • Protein Expression: CD8: Stably and predominantly expressed on cytotoxic T lymphocytes (CTLs). CD20: Exclusively expressed on B-lineage cells from pre-B to mature B cells and widely overexpressed on B-cell lymphomas, but absent on plasma cells. 
      • Signaling Pathway: CD8: Recruits Lck kinase upon TCR-MHC I binding to initiate cytotoxic activation cascades. CD20: Facilitates transmembrane calcium transport. CD8-directed engagement physically bridges cytotoxic T cells to eliminate CD20-expressing target B cells. 
      • Therapeutic Application: CD8 × CD20 platforms (such as in vivo CAR-T delivery vectors and bispecific antibodies) specifically redirect CD8+ T cells to eradicate CD20+ malignant B cells, driving potent anti-tumor efficacy with minimal off-target risks.
      CD8 Protein Expression in Spleen

      Strain specific CD8 expression analysis in homozygous B-hCD8/hCD20 mice by flow cytometry. Splenocytes were collected from homozygous B-hCD8/hCD20 mice (H/H,H/H) (female, 6-week-old), and analyzed by flow cytometry with species-specific anti-CD8 antibody (anti-human CD8A, Biolegend, 300908; anti-mouse CD8A, Biolegend, 100730; anti-human CD8B, BD, 742392).

      Strain specific CD8 expression analysis in homozygous B-hCD8/hCD20 mice by flow cytometry. Blood were collected from homozygous B-hCD8/hCD20 mice (H/H,H/H) (female, 6-week-old), and analyzed by flow cytometry with species-specific anti-CD8 antibody (anti-human CD8A, Biolegend, 300908; anti-mouse CD8A, Biolegend, 100730; anti-human CD8B, BD, 742392).
