Gene targeting strategy for B-NDG hTROP2 mice. The exon 1 of mouse Trop2 gene that encodes signal peptide, extracellular domain, transmembrane domain, cytoplasmic region are replaced by human counterparts in B-NDG hTROP2 mice. The promoter, 5’UTR and 3’UTR region of the mouse gene are retained. The human TROP2 expression is driven by endogenous mouse Trop2 promoter, while mouse Trop2 gene transcription and translation will be disrupted.
Species specific analysis of TROP2 gene expression in wild-type B-NDG mice and homozygous humanized B-NDG hTROP2 mice by RT-PCR. Skin was collected from wild-type B-NDG mice (+/+) and homozygous B-NDG hTROP2 mice (H/H). Mouse Trop2 mRNA was detectable in wild-type B-NDG mice but not in homozygous B-NDG hTROP2 mice. Human TROP2 mRNA was exclusively detectable in homozygous B-NDG hTROP2 mice but not in wild-type B-NDG mice.
Western blot analysis of TROP2 protein expression in homozygous B-NDG hTROP2 mice. Various tissue lysates were collected from wild-type B-NDG mice (+/+) and homozygous B-NDG hTROP2 mice (H/H), and then analyzed by western blot with anti-TROP2 antibody (Abcam, ab214488). 40 μg total proteins were loaded for western blotting analysis. TROP2 was detected in the lung, kidney and stomach of wild-type mice and B-NDG hTROP2 mice due to the cross-reactivity of antibodies, but not expressed in the heart, liver, spleen, brain, colon, testis and ovary.
