      Description
      • Background: ASGR1, a hepatocyte-enriched asialoglycoprotein receptor, recognizes desialylated glycoproteins bearing terminal galactose or GalNAc residues. Ligand binding triggers receptor-mediated endocytosis, followed by lysosomal trafficking of the ligands and recycling of the receptor to the cell surface.
      • Gene targeting strategy: The exons 1-9 of mouse Asgr1 gene that encode signal peptide, extracellular domain, transmembrane domain, cytoplasmic region and 3’UTR were knocked out.
      • Validation:

      Mouse Asgr1 mRNA was detectable only in wild-type C57BL/6JNifdc mice, but not in homozygous B-Asgr1 KO mice.

      Western blot: Asgr1 protein was only detected in the liver in C57BL/6JNifdc mice, but not in homozygous B-Asgr1 KO mice.

      mRNA Expression Analysis in B-Asgr1 KO Mice

      Species-specific analysis of Asgr1 gene expression in wild-type C57BL/6JNifdc mice and homozygous B-Asgr1 KO mice by RT-PCR. Liver was collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-Asgr1 KO mice (-/-). Mouse Asgr1 mRNA was detectable only in wild-type C57BL/6JNifdc mice, but not in homozygous B-Asgr1 KO mice.

      Protein Expression Analysis in B-Asgr1 KO Mice

      Western blot analysis of ASGR1 protein expression in wild-type C57BL/6JNifdc mice and B-Asgr1 KO mice. Liver tissue lysates were collected from C57BL/6JNifdc (+/+) and homozygous B-Asgr1 KO mice (-/-) (male, 6-9 weeks old), and analyzed by western blot with anti-Asgr1 antibody (Proteintech, 11739-1-AP). 40 μg of total protein was loaded for Western blotting analysis. Asgr1 was only detected in the liver of C57BL/6JNifdc mice, but not in homozygous B-Asgr1 KO mice.
